Flavobacterium difficile sp. nov., isolated from a freshwater waterfall

A bacterial strain designated KDG-16 T is isolated from a freshwater waterfall in Taiwan and characterized to determine its taxonomic affiliation. Cells of are Gram-stain-negative, strictly aerobic, motile by gliding, rod-shaped form light yellow colonies. Optimal growth occurs at 20–25 °C, pH 6–7, with 0% NaCl. Phylogenetic analyses based on 16S rRNA gene sequences an up-to-date core set reveal that affiliated species the genus Flavobacterium. Analysis shows shares highest similarity Flavobacterium terrigena DSM 17934 (97.7%). The average nucleotide identity, amino acid identity digital DNA–DNA hybridization values between closely related below cut-off 95–96, 90 70%, respectively, used for demarcation. Strain contains iso-C15:0, iso-C15:1 G iso-C17:0 3-OH as predominant fatty acids. polar lipid profile consists phosphatidylethanolamine, one uncharacterized aminophospholipid, phospholipid, two aminolipids lipids. major polyamine homospermidine. isoprenoid quinone MK-6. Genomic DNA + C content 31.6%. Based polyphasic data obtained, considered represent novel Flavobacterium, which name difficile sp. nov. proposed. type (= BCRC 81194 = LMG 31332 T).
